Background:Efficacy of Seroquel for major depressive disorder (MDD) has not been established. We sought to determine the effects of placebo on the risk of major depressive disorder (MDD) in adults. We compared the relative efficacy between placebo and a non-placebo regimen for MDD using the International Index of Depressive Disorder (IID) in the Primary Care Intervention (PCI) trial, a large randomized controlled trial of antidepressant treatment with quetiapine (Seroquel) and placebo in adults with MDD who were at high risk for the diagnosis of MDD. We also used the secondary outcome of suicide attempts in adults who had been taking quetiapine, as part of a larger, randomized controlled trial of antidepressant treatment with quetiapine and placebo in adults with MDD who had an initial diagnosis of MDD. The primary outcome of interest was the change from baseline to the end of follow-up in the number of suicide attempts in adults treated with quetiapine and placebo. We included adult participants who had been treated with quetiapine or placebo for at least three months and who met the criteria for a primary endpoint. We used intention-to-treat analysis to calculate the hazard ratio and 95% confidence interval. The primary endpoint was the risk of suicide attempts in adults treated with quetiapine and placebo compared with the placebo group at 1 year. The primary outcome was the change from baseline to the end of follow-up in the number of suicide attempts in adults treated with quetiapine and placebo compared with the placebo group at 1 year.
